Enzymatic hydrolysis of cuttlefish (Sepia officinalis) and sardine (Sardina pilchardus) viscera using commercial proteases: Effects on lipid distribution and amino acid composition ArchiMer

Total lipid and phospholipid recovery as well as amino acid quality and composition from cuttlefish (Sepia officinalis) and sardine (Sardina pilchardus) were compared. Enzymatic hydrolyses were performed using the three proteases Protamex, Alcalase, and Flavourzyme by the pH-stat method (24 h, pH 8, 50 degrees C). Three fractions were generated: an insoluble sludge, a soluble aqueous phase, and an oily phase. For each fraction, lipids, phospholipids, and proteins were quantified. Quantitative and qualitative analyses of the raw material and hydrolysates were performed. The main goal of this work was to up-grade sardine (Sardina pilchardus) by-products using mild procedure, environmentally sound, in order to obtain valuable compounds involving moderate investment and low energy consumption. Hydrolysis and ultrafiltration techniques have been investigated. Firstly, selected enzymes have been calibrated in order to determine their optimal conditions with a model substrate and to permit the comparison between them. Then, hydrolysis on sardine head and viscera have been carried out in two steps. The first one had the objective to determine the enzyme efficiency and the study area. The main objective of this study is the valorisation of by-products coming from processed tuna (Thunnus albacares). Therefore, enzymatic hydrolysis was selected, using an industrial large spectrum protease (Protamex). In a first phase, hydrolyses of these by-products (heads, tails, viscera) were carried out for long periods of time (12h) in order to establish the kinetics of the proteolysis of those different by-products and to obtain the approximate composition of the different resulting fractions and to determine the conditions which would allow the valorisation of these fractions. The cuttlefish Sepia officinalis, is one of the main species capted in the Basse-Normandie fisheries. These animals are principally exportated in eviscerated and frozen form to mediteranean countries and to Japan. Then, high quantities of co-products are available from factories transformation of cephalopods. Enzymological and biochemical studies on co-products demonstrated an elevated potential in the valorisation of viscera by silage technical. Two silages are developed in our laboratory: LBBMA4 and LBBMA25. Primarly, they are studed in vitro as substrate for the culture of micro-organisms.
